Is it possible to control VDJ via a mixer that I have connected to my soundcard?
Secondly, do I need to 2 x line-in's to effectively connect my DJM600 to my computer?

You can controll VDJ with midi mixers only, connected with USB cable to computer.
DJM600 of Pioneer is standard analog mixer that need 2 sreteo outs for separane decks. So you will need 2 streo outputs.
